The image on the cover is a photograph by the American artist David Wojnarowicz, depicting how Indians hunted buffalo by causing them to run off cliffs. Wojnarowicz identifies himself and ourselves with the buffalo, pushed into the unknown by forces we cannot control or even understand. Wojnarowicz is an activist artist and writer whose work has created controversy recently through its uncompromising depiction of the artist's homosexuality, his infection by the H.I.V. virus and the political crisis surrounding AIDS.

O U2 reverteu os royalties da venda desse single, para a campanha na luta contra a Aids.
O Lado B desse single é uma cover de Satelitte Of Love, do Lou Reed.
O videoclipe de One tem três versões diferentes: um videoclipe que sugere que "One" se trata de um filho gay que está confessando ao pai que é HIV positivo. Filmado em Berlin, mostra os integrantes da banda vestidos de drag queens e Bono cantando a canção para seu pai ( representado pelo próprio Bob Hewson, pai de Bono).
O outro vídeo mostra flores, búfalos e a palavra "one" escrita em diversas linguagens diferentes. A capa do single de One que mostra esses búfalos, foi fotografada pelo artista David Wojnarowicz, que morreu de Aids.
Outro vídeo mostra Bono em um bar, fumando um More mentolado e bebendo uisque e cerveja Guinness. Mostra a clássica cena em que Bono canta o trecho "We're one, but we're not the same. Well, we hurt each other, then we do it again.You say......." e pára de cantar, olhando fixo para a câmera.
Existem interpretações diferentes do significado da canção "One". Ela pode representar:
1. O retrato da amargura própria do fim de um grande amor: One love, we get to share it. Leaves you baby, if you don't care for it. Did I disappoint you? Or leave a bad taste in your mouth?
2. O diálogo entre um pai e um filho que está prestes a morrer de Aids e quer ter uma proximidade que nunca teve junto a esse pai (versão defendida com base na dedicatória feita no álbum): Have you come here for forgiveness? Did I ask too much, more than a lot? You gave me nothing, now it's all I got
3. A relação vivida entre os membros da banda, que na altura da gravação do álbum era tensa (versão baseada em fatos reais): We're one, but we're not the same. We get to carry each other, carry each other
4. A relação entre os povos das duas Alemanhas depois da queda do muro de Berlim (versão baseada no contexto histórico e local da gravação do álbum, Berlim): we're one but we're not the same
5. A relação com Deus, num misto de desapontamento e busca de apoio (versão sustentada pela espiritualidade da banda)
6. A relação entre os países privilegiados/desafortunados (versão sustentada pelo ativismo da banda, particularmente a africanidade): one life with each other, sisters, brothers.
Eleita a música da década por várias revistas, é uma letra com inúmeras interpretações: há quem considere apenas uma linda letra de amor; há quem considere uma letra de ódio. Bono disse que a letra poderia significar um fictício diálogo entre Judas e Jesus Cristo, como também uma conversa entre um filho aidético pedindo ajuda ao pai. Em todos os casos, é um dos momentos mais pungentes da carreira do grupo.
